So three options.
first would be cortana could be disabled, which can be fixed by enabling her,
second could be a microphone issue like deadly pants said
third if you havent restarted your computer at all when this issue started, someone could have ended her process in task manager but thats really all i can think of

Why do you want to use cortana in the first place? Check your mic first, see if it records any audio.
