Can I leave the existing DIMMs that came with the system (2x4G) and add an additional 2x16G DIMMS for a total of 40G?
Do I need to place all the DIMMS in matched pairs of slots?. For example the 4G DIMMs in slots 1 & 3 and the 16G DIMMs in slots 2 & 4 or can the DIMMS go in any slots (4G DIMMs in slots 1 &2 and 16G DIMMS in slots 3&4)?

Are you sure that's an optimum/ideal/recommended configuration?
Depending on how the northbridge/ram slots are configured, you might suffer from not having the same size of ram stick in each slot.
I've been looking to see if this is addressed rigorously anyware, and so far, no luck.
In this case one could try benchmarking it to see if you see any noticeable differences:
(1) 2 x 16
(2) 2 x 16 + 2 x 4
(3) 4 x 8
You can now buy new 16GB RAM for £69, so if there really is a need to upgrade the RAM on a £2k system, it seems best to do it thoroughly, especially since it's cheap to do it.
As in go to 32GB by 4 x 8GB, or else to 64GB by 4 x 16GB.
I think 2 x 16 + 2 x 4 is a potentially half-a&&ed measure.
If you can actually SEE any benefit from the extra 8GB (from the OEM 2 x 4GB) on top of the 32GB, then you should really go whole hog to 64GB (4 x 16GB).
