Computer won't start, circle with diagonal line comes on?

When I turn on my computer the Apple logo comes on and then the wheel starts spinning. After about a minute the Apple logo is replaced by a circle with a diagonal line ( as in street signs do not enter). The wheel still spins. Can you help?

f you're using Mac OS X 10.2 or later, you can start up your computer in Safe Mode, which includes an automatic disk check and repair. Hold down the shift key on start up. If this works follow these procedures:
